    Chromosomal anchoring of linkage groups and identification of wing size QTL using markers and FISH probes derived from microdissected chromosomes in Nasonia(Pteromalidae : Hymenoptera)

    Nasonia vitripennis is a small parasitic hymenopteran with a 50-year history of genetic work including linkage mapping with mutant and molecular markers. For the first time we are now able to anchor linkage groups to specific chromosomes. Two linkage maps based on a hybrid cross (N. vitripennis x N. longicornis) were constructed using STS, RAPID and microsatellite markers, where 17 of the linked STS markers were developed from single microdissected banded chromosomes. Based on these microdissections we anchored all linkage groups to the five chromosomes of N. vitripennis. We also verified the chromosomal specificity of the microdissection through in situ hybridization and linkage analyses. This information and technique will allow us in the future to locate genes or QTL detected in different mapping populations efficiently and fast on homologous chromosomes or even chromosomal regions. To test this approach we asked whether QTL responsible for the wing size in two different hybrid crosses (N. vitripennis x N. longicornis and N. vitripennis x N. giraulti) map to the same location. One QTL with a major effect was found to map to the centromere region of chromosome 3 in both crosses. This could indicate that indeed the same gene/s is involved in the reduction of wing in N. vitripennis and N. longicornis. Copyright (C) 2003 S. Karger AG, Basel

    Glass transition in Ultrathin Polymer Films : A Thermal Expansion Study

    Glass transition process gets affected in ultrathin films having thickness comparable to the size of the molecules. We observe systematic broadening of glass transition temperature (Tg) as the thickness of the polymer film reduces below the radius of gyration but the change in the average Tg was found to be very small. Existence of reversible negative and positive thermal expansion below and above Tg increased the sensitivity of our thickness measurements performed using energy dispersive x-ray reflectivity. A simple model of Tg variation as a function of depth expected from sliding motion could explain the results. We observe clear glass transition even for 4 nm polystyrene film that was predicted to be absent from ellipsometry measurements of thicker films.Comment: 11 pages, 5 figure

    Response of Root Properties to Tripartite Symbiosis between Lucerne (Medicago sativa L.), Rhizobia and Mycorrhiza Under Dry Organic Farming Conditions

    It is generally considered that root turnover is a major contributor to organic matter and mineral nutrient cycles in organic managed agroecosystems. Approach: This study designed to investigate whether microbial activity could affect on root properties of lucerne in an organically managed field under dry weather conditions. The trial was laid out as a factorial experiment in the fields of the University of Natural Resources and Applied Life Sciences, Vienna-Austria at Raasdorf in 2007. The experimental factors of Sinorhizobium meliloti and arbuscular mycorrhiza (AM) including Glomus etunicatum, G. intraradices and G. claroideum and irrigation levels were tested. Results: Results showed that increasing water deficit affected root dry weigh, specific root mass and root length significantly at 1% level and co-inoculation of rhizobium and mycorrhiza with irrigation could increase all root parameters. Data’s of variance analysis for mycorrhizal colonization showed that main effect of using mycorrhiza had significant effects on root parameters at 5% and 1% probability level in first and second harvest, respectively. Results of mean comparisons by Duncan’s multiple range test showed that mycorrhizal colonization was higher in the inoculated treatments by rhizobium , mycorrhiza and irrigated plots in both harvests. Double interaction of mycorrhiza and irrigation was higher in both harvests (37.05% and 65.73%, respectively). Conclusion: Hence, it can be suggested that the tripartite symbiosis of S. meliloti, AM and lucerne can improve the performance of lucerne in organic farming and under dry conditions. Such traits could be incorporated into breeding programs to improve drought tolerance especially in organic fields

    Genetic diversity and distance among Iranian and European alfalfa (Medicago sativa L.) genotypes

    Alfalfa is the best known fodder crop with high ability of biological nitrogen fixation and drought tolerance in dry, Pannonian region of east Austria. Different morphological and physiological characteristics of 18 alfalfa genotypes from different geographical origins, 8 Iranian ecotypes and 10 European cultivars were evaluated under irrigated and rainfed conditions during 2006-08 cropping seasons. The objectives of this study were to measure genetic distance and divergence among genotypes and to classify them based on morphological and physiological characters. Cluster analysis differentiated Iranian ecotypes and European cultivars from each other under irrigated condition, and when data averaged across two environments (irrigated and rainfed). However, under rainfed conditions small changes occurred in grouping of genotypes due mainly to differential responses of the genotypes to rainfed condition. Considerable genetic distance observed between Iranian and European genotypes. Different crossing programs are recommended between Iranian and European genotypes to develop new alfalfa cultivars

    Plant biomass nitrogen and effects on the risk of nitrate leaching of intercrops under organic farming in Eastern Austria

    Data on the potential of intercrops to reduce soil nitrate contents, on their N accumulation and biological nitrogen fixation (BNF) are lacking for organic farming in the dry, pannonic region of Eastern Austria. The effect of legumes, non-legumes, and legumes + non-legumes used as intercrops on inorganic soil nitrogen, crop yield and biomass N, and BNF were tested in comparison to bare fallow. Non-legumes and legumes + non-legumes were more efficient than legumes in reducing inorganic soil N contents in autumn and nitrate contents in soil solution from the subsoil in winter. This reduction in inorganic soil N did not last until March of the following year due to an N mineralisation from the mulch. The legume + non-legume mixture contained a larger amount of crop N than both legumes and non-legumes. This was due to the combined effect of soil-N uptake by the non-legumes and BNF by the legumes in the mixture

    Deep XMM-Newton observation of a northern LMC field: I. Selected X-ray sources

    First results from a deep XMM-Newton observation of a field in the Large Magellanic Cloud (LMC) near the northern rim of the supergiant shell LMC 4 are presented. Spectral and temporal analyses of a sample of selected X-ray sources yielded two new candidates for supernova remnants, a supersoft X-ray source and a likely high mass X-ray binary (HMXB) pulsar. From the fourteen brightest sources up to ten are active galactic nuclei in the background of the galaxy which can be used as probes for the interstellar medium in the LMC. From the three previously known HMXBs the Be/X-ray binary EXO 053109-6609.2 was the brightest source in the field, allowing a more detailed analysis of its X-ray spectrum and pulse profile. During the pulse EXO 053109-6609.2 shows eclipses of the X-ray emitting areas with increased photo-electric absorption before and after the eclipse. The detection of X-ray pulsations with a period of 69.2 s is confirmed for RX J0529.8-6556 and a possible period of 272 s is discovered from XMMU J053011.2-655122. The results are discussed with respect to individual sources as well as in the view of source population studies in the vicinity of the supergiant shell LMC 4

    Changes in olfactory bulb volume following lateralized olfactory training.

    Repeated exposure to odors modifies olfactory function. Consequently, "olfactory training" plays a significant role in hyposmia treatment. In addition, numerous studies show that the olfactory bulb (OB) volume changes in disorders associated with olfactory dysfunction. Aim of this study was to investigate whether and how olfactory bulb volume changes in relation to lateralized olfactory training in healthy people. Over a period of 4 months, 97 healthy participants (63 females and 34 males, mean age: 23.74 ± 4.16 years, age range: 19-43 years) performed olfactory training by exposing the same nostril twice a day to 4 odors (lemon, rose, eucalyptus and cloves) while closing the other nostril. Before and after olfactory training, magnetic resonance imaging (MRI) scans were performed to measure OB volume. Furthermore, participants underwent lateralized odor threshold and odor identification testing using the "Sniffin' Sticks" test battery.OB volume increased significantly after olfactory training (11.3 % and 13.1 % respectively) for both trained and untrained nostril. No significant effects of sex, duration and frequency of training or age of the subjects were seen. Interestingly, PEA odor thresholds worsened after training, while olfactory identification remained unchanged.These data show for the first time in humans that olfactory training may involve top-down process, which ultimately lead to a bilateral increase in olfactory bulb volume

    Suitability of drought tolerance indices for selecting alfalfa (Medicago sativa L.) genotypes under organic farming in Austria

    In eastern Austria, alfalfa is usually grown as a rainfed crop in crop rotations in organic farming systems, where year-to-year rainfall fluctuations cause different levels of drought stress. To identify the suitability of different alfalfa genotypes and drought tolerance indices, 18 contrasting alfalfa genotypes were evaluated under irrigated and rainfed conditions at the research station of the University of Natural Resources and Life Sciences (BOKU), Vienna, Austria, during 2006-08. The first study year (2006) was considered as the establishment year. Five drought tolerance selection indices were estimated based on shoot dry matter, total biomass yield and biological nitrogen fixation (BNF) data. The correlation between irrigated and rainfed performances increased (from r=-0.17 to 0.56) with decreasing stress intensity from the first to the second year. Genotypes Sitel, Plato ZS, Vlasta and NS-Banat were the best genotypes based on their performance under both conditions. Drought tolerance selection indices TOL and SSI showed high correlations (r = 0.32 to 0.81) only with rainfed performance, and SSI was the index that best identified genotypes with high yield potential under rainfed conditions. Indices STI and GMP were the ones that best identified genotypes with high performance under both conditions
